Adamax (N-Acetyl Semax Adamantane, Ac-MEHFPGP-AG-NH₂) is a synthetic heptapeptide with CAS number 114681-65-1, molecular formula C₅₀H₆₉N₁₁O₁₁S, and a molecular weight of approximately 1,032.23 Da. Adamax is a chemistry-modified derivative of Semax (a peptide on the ACTH 4-10 sequence) featuring dual modifications: an N-terminal acetyl group that enhances resistance to aminopeptidase degradation and improves blood-brain barrier (BBB) permeability in research models, and a C-terminal adamantane moiety — a rigid, lipophilic tricyclic cage structure — that further increases lipophilicity, enzymatic stability, and CNS uptake observed in vitro.

In laboratory research, Adamax is studied for its modulation of brain-derived neurotrophic factor (BDNF) expression, receptor phosphorylation and sensitivity in hippocampal models, melanocortin receptor subtype (MC3R, MC4R) interactions, and monoamine neurotransmitter metabolism (dopamine, norepinephrine, serotonin pathways). Researchers employ it in neuronal cell culture assays, CREB phosphorylation signaling studies, oxidative stress-induced neuronal damage models, and comparative in-vitro structure–activity studies with unmodified Semax to evaluate the structural effects of the adamantane modification.
