Cagrilintide is a synthetic 37-amino acid research peptide (CAS 1415456-99-3, molecular formula C₁₉₄H₃₁₂N₅₄O₅₉S₂, MW ~4,409 Da) constructed on a native human amylin (islet amyloid polypeptide, IAPP) backbone. The peptide retains the Cys2–Cys7 disulfide bridge of native amylin and incorporates three residue substitutions — N14E, V17R, and P37Y — along with an N-terminal C20 eicosanedioic fatty diacid conjugated via a γ-glutamic acid spacer enabling in vitro albumin-binding behavior.
In laboratory research, Cagrilintide is utilized in amylin receptor complex binding studies across AMY1 (CTR + RAMP1), AMY2 (CTR + RAMP2), and AMY3 (CTR + RAMP3), calcitonin receptor (CTR) binding characterization, Gs-coupled cAMP accumulation assays (reported EC50 ~49 pM at hAMY3R, ~62 pM at hCTR), receptor-binding affinity determinations (Ki ~3.5 nM at hAMY3R, ~6.8 nM at hCTR), and comparative in vitro structure–activity studies against other amylin-backbone research peptides.
