Description

PP-3R is a synthetic 39-amino acid research peptide (CAS 2381089-83-2, molecular formula C₂₂₁H₃₄₂N₄₆O₆₈) constructed on a GIP (glucose-dependent insulinotropic polypeptide) backbone with in vitro binding activity characterized at the glucagon-like peptide-1 receptor (GLP-1R), glucose-dependent insulinotropic polypeptide receptor (GIPR), and glucagon receptor (GCGR). The peptide incorporates three non-coded amino acid residues: α-aminoisobutyric acid (Aib) at positions 2 and 20 for resistance to dipeptidyl peptidase-IV (DPP-IV) enzymatic degradation, and α-methyl-L-leucine (αMeL) at position 13 for optimized in vitro binding at GIPR and GCGR. A C20 fatty diacid moiety is conjugated via a linker at Lys17 to enable non-covalent in vitro albumin binding.

In laboratory research, PP-3R is used in multi-receptor binding-affinity studies, GLP-1R/GIPR/GCGR cellular signaling crosstalk investigations, cAMP accumulation assays across receptor-expressing cell lines, cellular energy-metabolism modulation experiments in cell-culture models, metabolic signaling pathway studies, and comparative in vitro structure–activity studies against multi-receptor and mono-receptor research-grade reference peptides. Cryo-EM structural studies have characterized distinct binding poses of PP-3R at each target receptor, revealing receptor-specific ECL1 recognition mechanisms.
