Description

PP-2T is a synthetic 39-amino acid research peptide (CAS 2023788-19-2, molecular formula C₂₂₅H₃₄₈N₄₈O₆₈, MW ~4,813.45 Da) constructed on a native GIP (glucose-dependent insulinotropic polypeptide) backbone. The peptide features two non-natural α-aminoisobutyric acid (Aib) residues at positions 2 and 13 for dipeptidyl peptidase-IV (DPP-IV) resistance, and a C20 fatty diacid (eicosanedioic acid) conjugated to Lys20 via a glutamic acid and dual AEEA (aminoethoxyethoxyacetic acid) linker enabling in vitro albumin-binding behavior. In cellular assays, PP-2T exhibits imbalanced binding affinity favoring GIPR over GLP-1R and demonstrates biased signaling toward cAMP generation over β-arrestin recruitment at GLP-1R.

In laboratory research, PP-2T is utilized in dual-receptor cAMP accumulation assays, biased-signaling characterization at GLP-1R, GIPR-mediated cAMP pathway activation studies, branched-chain amino acid (BCAA) catabolism investigations in brown adipose tissue cell models, comparative in-vitro structure–activity studies versus single-receptor (GLP-1R-only) research peptides, and Akt/mTOR signaling pathway research.
