NAD+ (Nicotinamide Adenine Dinucleotide) is the oxidized form of a vital dinucleotide coenzyme present in all living cells, with the molecular formula C₂₁H₂₇N₇O₁₄P₂, a molecular weight of approximately 663.43 Da, and CAS number 53-84-9. As a central mediator in cellular redox biochemistry, NAD+ participates in over 500 known enzymatic reactions, functioning as a primary electron carrier in glycolysis, the tricarboxylic acid (TCA) cycle, and the mitochondrial electron transport chain. Beyond its role as a coenzyme, NAD+ serves as a consumed substrate for three major enzyme families: sirtuins (SIRT1–SIRT7, class III histone deacetylases), poly(ADP-ribose) polymerases (PARPs, involved in DNA damage repair signaling), and cyclic ADP-ribose synthases (CD38/CD157 ectoenzymes).

In laboratory research, NAD+ is utilized in sirtuin deacetylase activity assays, PARP enzymatic activity measurements, CD38 ectoenzyme kinetic studies, mitochondrial electron transport chain functional assays, and cellular metabolic flux analysis using isotope tracing. Researchers also employ NAD+ in studies of NAD+-dependent calcium signaling via cADPR production, epigenetic regulation through histone deacetylation, and age-related changes in the NAD+ metabolome.
