Description

PP-1S is a synthetic 31-amino acid research peptide with molecular formula C₁₈₇H₂₉₁N₄₅O₅₉, a molecular weight of approximately 4,113.58 Da, and CAS number 910463-68-2. The peptide shares 94% structural homology with native human glucagon-like peptide-1 (7-37) and incorporates three modifications: an α-aminoisobutyric acid (Aib) substitution at position 8 for dipeptidyl peptidase-IV (DPP-IV) resistance, an Arg substitution at position 34 to ensure single-site fatty acid conjugation, and a C18 octadecanedioic fatty diacid moiety conjugated at Lys26 via a γ-glutamic acid spacer and mini-PEG (aminoethyloxy-acetyl) linker enabling in vitro albumin-binding behavior.

In laboratory research, PP-1S is used in GLP-1 receptor–binding studies via cAMP/PKA and β-arrestin signaling pathway assays, in vitro pancreatic beta-cell model assays, cellular signaling experiments in CNS cell culture systems, receptor-binding affinity characterizations (Ki ~0.38 nM), and comparative in vitro structure–activity studies against other research-grade receptor-binding peptides.
