5-Amino-1MQ (5-Amino-1-methylquinolinium) is a small molecule quinolinium compound with the molecular formula C₁₀H₁₁N₂⁺ (supplied as the iodide salt, C₁₀H₁₁IN₂, MW ~286.11 Da) and CAS number 42464-96-0. 5-Amino-1MQ is a substrate-site-targeting, selective inhibitor of nicotinamide N-methyltransferase (NNMT) with an IC₅₀ of 1.2 μM (at 50 μM SAM, 100 μM nicotinamide). NNMT catalyzes the N-methylation of nicotinamide using S-adenosylmethionine (SAM) as the methyl donor, producing 1-methylnicotinamide (1-MNA) and S-adenosylhomocysteine (SAH). By inhibiting NNMT, 5-Amino-1MQ modulates the cellular SAM/SAH methyl donor ratio and sirtuin-1 (SIRT1) activity.

In laboratory research, this compound is utilized in 3T3-L1 adipocyte differentiation assays (EC₅₀ ~30 μM for lipid accumulation reduction), 1-MNA level measurements in cell culture models (EC₅₀ ~2.3 μM), myoblast differentiation and muscle stem cell fusion studies, metabolic flux analysis, and epigenetic methylation dynamics investigations. Notably, 5-Amino-1MQ demonstrates selectivity for NNMT without affecting related methyltransferases or other enzymes in the methyltransferase family.
