Description

SS-31, also known as elamipretide (synonyms MTP-131, Bendavia; sequence H-D-Arg-Dmt-Lys-Phe-NH₂, where Dmt = 2′,6′-dimethyl-L-tyrosine), is a synthetic aromatic-cationic Szeto-Schiller mitochondria-targeting tetrapeptide. Molecular formula C₃₂H₄₉N₉O₅ (free base), molecular weight ~639.79 Da, CAS 736992-21-5 (free base; acetate salt 1334953-95-5). It is a non-proteinogenic peptide containing a D-arginine residue and an unnatural dimethyltyrosine residue with a C-terminal primary amide, studied for its potential-independent partitioning into the inner mitochondrial membrane and association with the phospholipid cardiolipin.

In laboratory research, SS-31 is utilized in in-vitro studies of mitochondrial bioenergetics (e.g., oxygen-consumption-rate / Seahorse-style respiration assays), electron-transport-chain coupling and respiratory-supercomplex assembly investigations (e.g., BN-PAGE), cardiolipin-binding and lipid-interaction biophysical assays, reactive-oxygen-species and oxidative-stress assay development in cultured cells (e.g., MitoSOX-type readouts), mitochondrial-membrane-potential and cristae-ultrastructure morphology studies (e.g., TMRM/JC-1, electron/fluorescence microscopy), and as a reference standard for HPLC and LC-MS analytical method development.
